Is it possible to upgrade directory from 4.1 to 4.3, or do we need to upgrade to 4.2 first?

You only need to update the database
Run upgrade script 4.1 to 4.2 and then 4.2 to 4.3
Obviously take a backup of v4.1 before you start in case there are problems
Then use the latest version of the website v4.3
